Title: Exploiting Bioorthogonal Chemistry to Modulate the Photophysical and Photochemical Properties of Transition Metal Complexes for Bioimaging and Photocytotoxic Applications

Abstract: Bioorthogonal chemistry has emerged as a versatile approach to examine the localization, dynamics, and functions of biomolecules in their native environments. It also serves as a powerful pretargeting strategy for molecular imaging and drug delivery. With our ongoing interest in the design of photofunctional transition metal complexes as bioimaging reagents and photocytotoxic agents, we believe that the strategic coupling of bioorthogonal functionalities with photoactive transition metal complexes will lead to the development of innovative theranostic agents. In this seminar, I will describe our recent designs of photofunctional rhenium(I), ruthenium(II), and iridium(III) polypyridine complexes as activatable bioorthogonal reagents. These complexes not only display phosphorogenic properties (i.e., phosphorescence turn-on) upon the bioorthogonal reaction, but also enhanced singlet oxygen photosensitization, thereby enabling controllable photocytotoxicity. The focus will be on the photophysical and photochemical properties, bioorthogonal reactivity, cellular uptake and localization, and (photo)cytotoxic activity of these photofunctional transition metal complexes.
